It is possible to do things while a backup is being made. You will notice that both activities will take longer as you are dividing your resources between two tasks. The backup task is more resource intensive so what you are doing may be much slower to happen.
If you turn the power off nothing can happen. If you log off then the computer can do things. When I'm going to run a backup after completing my day I hit CTRL+ALT+DELETE then ENTER and lock the computer. That way the system has all of its resources available to do whet needs doing and nobody will come in and play. That also lets me set the backup program's little slid to 100% when it asks about resource sharing. All resources are devoted to the backup.
Works for me but not set in stone.

In addition to that you'll want to ensure power management is not enabled. If the system goes into sleep it may prevent the backup from starting or cut it off while its working. Also take note actions such as purges or restores are not automated and require you to actually start them via the web interface. Once one of those is ongoing it will continue to process unless you terminate it.
