I have Norton Internet Security 2012 ver 19.7.0.9.
I use Outlook 2007 on my desktop with a corporate Exchange 2010 server.
I very much doubt that it's related to my ISP as I have recently switched over from using Kaspersky and I never had such problems. So the infrastructure is the same but the protection layer has changed.
Are there any settings to be checked?

As Phil_D said, this SMTP error most likely indicates a problem at the email server. However, to make sure that Norton isn't causing a problem in sending, you can disable Outgoing Email Scanning in Settings > Network > Message Protection > Email Antivirus Scan Configure[+]. Norton will alert you that email scanning is turned off and will want to fix it. By hovering your mouse cursor over the words "Email Protection" on the main Norton Advanced screen and clicking "Ignore" in the popup, you can turn off the alerts. I would first try resending your messages, as this may be only a temporary email server problem.
SendOfJive provided excellent diagnostic advice which may help narrow down the issue.
Since you mentioned having recently switched from Kaspersky, I have two thoughts.

After a day or so w/o any problems, I turned on again the outgoing email protection, and soon enough the unsent emails and above errors - returned....
Just leave Outgoing Email Scanning disabled, then. It does nothing to protect your computer, anyway. It simply scans outgoing messages to check that you are not sending any infected attachments out. Since you are running Norton, it is not likely that there will be any malware on your machine to begin with, and, secondly, your recipients are almost certainly protected by their own antivirus programs. Outgoing scans are completely expendable.

But I do nitoce that quite a few emails I try to send from my default (private POP3) account - are not getting sent.
I see the supposedly sent email in the "sent items" folder - but the recipient does not receive them - and there's no error message.
This happened to me tonight with 4 emails (2 separate recipients, one of the a BB user and the other an Exchange 2010 user).
Very strange.
I would certainly check with your ISP, if you have not done so already.
I don't think the ISP is handling the mail, it's just providing the internet connection. It's the company exchange server working as the email provider.

I do have an ISP which provides me the POP3 account, while my workplace provides the Exchange 2010 (server and) account.
On my PC I use Outlook 2007, where I keep all read POP3 emails (they are deleted from the server), while the Exchange emails are kept on the Exchange server (and a copy on my home PC).
Yesterday I checked my Outlook 2007 settings on my PC and the default email address was the POP3, since I don't want my personal emails sent from home going through the Exchange account.
AFAIK This problem only occurs when I try to send (or reply to) emails from my PC using the POP3 account. If I change the account to the Exchange - I have not noticed any issues.
A partial problem ossurs on my iPhone/iPad2 when replying to emails received to my POP3 account (no such issue with my Gmail/Exchange accounts). I don't think I have problems when composing messages, just the replies.
