Is the standard version of NIS09 better than the OEM version that came preinstalled on my HP laptop? I extended the subscription with the 3-user NIS09 key from my previous laptop. I also have all of the updates from Live Update. Are there any significant differences between the two besides the "About" box listing HP as the OEM partner.
There is no difference in features or protection that is provided by the Norton program. The only difference is with the initial subscription, and you may receive future upgrades for OEM version only after renewing subscription. Once you activate it using your NIS 2009 Retail version (Standard) Product Key, it gets subscription of that and you will be getting future upgrades/updates without any problem.

When it's time to upgrade to NIS10, will NIS10 uninstall the OEM version of NIS09 and install the regular version of NIS10? As a reminder, I already have a regular 3-user subscription.
Yes, since you have now activated the OEM version with the Product key of regular retail version.
