Norton Internet Security 2011 is designed in such a way that the email client does not have both the Norton AntiSpam folder and the Junk folder. If the Junk folder is present in the email client, then NIS 2011 uses Junk folder to collect the spam mails. If Junk folder is not present, then NIS 2011 created the Norton AntiSpam folder and moves the spam mails to the Norton AntiSpam folder. So please check your Junk folder to see whether the spam mails are moving to it.
This does not apply if you are updating from 2010 version to 2011.
I also have the same problem, the Antispam buttons in the tool bar are their ONLY until you move the mouse close to it then it goes out (greyed ou) & you can not use it at all. I checked all three machines I have & it (antispam) doesn't work on any of the 3 machines. If you right click on a email message, their is NO antispam message available like it used to be years ago.
1) Allows using the option for sending the misclassified spam emails for analysis to Symantec, i.e., mails that are not tagged spam but are found to be spam by the user.
2) Allows adding the sender's email address for the misclassified email to the Blocked List
3) Moves the misclassified email from a non-junk email folder to the Junk/Norton AntiSpam folder whichever is the spam email destination
Hence the "This is Spam" button is expected to be enabled only for emails in the non-junk email folders with atleast one email present in them.
For your second issue of the button not being enabled for Norton AntiSpam tagged emails, were the emails present in Junk E-mail folder(which is where the tagged emails are expected to move)?
If so and since the email has already been identified by Norton as Spam, there would be no necessity for the button to be enabled. Hence, "This is Not Spam" alone would be enabled for emails in the Junk E-mail folder.
For the first issue,the Norton Anti-Spam folder would get created if a Junk folder is not detected within your Outlook data folder.
From my previous installations of Outlook 2007 and NIS 2010 I still have a Norton Spam Folder (now empty of course).
Is it safe to now delete this folder ?

I would not remove it. As surya mentioned if you upgrade from 2010 and already have a Norton Antispam folder it will remain there.
I also have Outlook 2007 with a Norton Antispam folder and all of my Norton identified spam email still gets moved to that folder and not the Junk folder.
If you want to confirm for sure which folder is being used by NIS you could always temporarily add your own email address to the Blocked list and then send yourself a test email and see which folder it gets moved to.
Of course don't forget to add yourself back to the Allowed list once you are done testing.
