Windows Defender and Vista User Account Control

Should I have these 2 programs on or off? I used to have windows defender on, with no trouble, but I read on these forums that it should be off. So I turned it off. I still have Vista User Account Control on. I like to have it on as an extra  safety step, and also with 3 kids, it should prevent them from downloading unwanted programs, since only I have the password. Once in a while when I am on a standard account and I click on NIS settings It asks for my UAC password and when I click cancel I get the familiar internal program error 40028 error code 0x80004005 instead of access denied, you have to be the administrator to perform this action. But I can live with that  for the extra safety of the UAC.  So I would rather not turn off the UAC.  Does anybody have either or both on with no conflicts?
